Clinical Research
- Phase 3 Trial: Metformin for Metabolic Support During ADT
A correlative analysis from the PRIME phase III randomized trial suggests that metformin may help mitigate harmful metabolic and hormonal changes caused by androgen deprivation therapy (ADT) in men with prostate cancer. The study found that metformin was particularly effective at offsetting the rise in leptin, a hormone linked to insulin resistance and potentially aggressive cancer biology, while also positively influencing other biomarkers like IL-6 and C-peptide. Although the results do not yet prove that metformin improves survival, they support its use as a supportive therapy to reduce the metabolic burden, such as weight gain and insulin resistance, in patients receiving hormone treatment.
- Phase 1/2 Trial: CRISPR-Edited Regulatory T Cells (CRG-150)
The CRG-150 program has launched a first-in-human testing phase to evaluate a CRISPR-edited regulatory T cell (Treg) therapy for solid tumors, including metastatic prostate cancer. This innovative approach differs from traditional cell therapies by reprogramming Tregs, which typically suppress immune responses, to instead support anti-tumor activity and reshape the suppressive tumor microenvironment. The trial’s initial focus is to ensure the therapy can be manufactured reliably and administered safely without causing unacceptable immune toxicity.
- Phase 1/2 Trial: Clinical Progress of TACC3 Inhibitor AO-252
Ongoing clinical updates for AO-252, a first-in-class TACC3 inhibitor, have demonstrated an 80% clinical benefit rate in heavily pre-treated patients when using a twice-daily dosing schedule. The therapy features a unique dual mechanism that combines direct cytotoxic effects with the activation of the cGAS/STING pathway, potentially enhancing immune responses against the tumor. Development is now shifting to an expansion phase involving a new lipid-based softgel formulation designed to improve drug absorption and consistency as it enters targeted testing for prostate and ovarian cancers.

- Fully Human Monoclonal Antibody Targeting TβRI
A novel, fully human monoclonal antibody developed by Umeå University has shown the ability to halt both tumor growth and metastatic spread in aggressive preclinical prostate cancer models. By specifically blocking oncogenic signaling through the TGF‑β receptor I (TβRI), the antibody prevents the downstream effects that drive invasion and the formation of metastatic niches. Preclinical safety data are encouraging, as the antibody did not cause adverse effects on body weight or heart function, suggesting it may avoid the systemic toxicity often associated with broader TGF-β pathway inhibition.
- Sonodynamic Nanotherapy for Bone Metastases
A new therapeutic strategy called sonodynamic nanotherapy uses the PN20@Rx nanoparticle system to specifically target and destroy bone metastases. These nanoparticles are guided to metastatic sites by alendronate and, once activated by ultrasound, produce reactive molecules that damage cancer cells while weakening their antioxidant defenses. In addition to direct cell killing, this method stimulates the immune system by reducing proteins that help cancer cells evade detection, and animal models have shown it can clear bone tumors while limiting further spread to the lungs.
