How DNA “Switches” in Prostate Cancer May Open New Treatment Paths
Researchers at USC have found that prostate cancer cells may rely on a 3D network of DNA “switches”,called enhancers, to keep cancer genes turned on. Enhancers are not genes themselves; they are stretches of DNA that help control when nearby or distant genes are active and how strongly they are expressed.
In this study, the team analyzed 201 datasets from prostate tumors and healthy prostate tissue and identified 3,216 prostate-cancer-specific enhancers. They then used a high-resolution mapping method to show that these enhancers form an interconnected network in cancer cells, but not in normal prostate cells.
The key finding was that some enhancers acted like central hubs. When the researchers deleted those hubs with CRISPR, the whole network became disrupted, cancer-promoting genes were switched off, and cancer cell growth slowed. That suggests future therapies may not need to target each bad gene one by one, but could instead target the regulatory control points that drive many genes at once.
